Caffeine Protects the Brain From Stress: Scientists Reveal How Coffee Affects Inflammation and Mood

A series of neurobiological studies shows that moderate caffeine consumption can curb inflammatory processes in nervous tissue and reduce the risk of depressive symptoms. According to a scientific review published in Translational Psychiatry, caffeine blocks adenosine receptors and regulates neurotransmitter metabolism, though excessive doses have the opposite effect, increasing anxiety.
Caffeine’s biological mechanism goes far beyond merely boosting wakefulness.
Adenosine Blockade and Protecting Neurons From Inflammation
The alkaloid primarily targets A1 and A2A adenosine receptors. By temporarily neutralizing drowsiness-inducing adenosine molecules, caffeine simultaneously stimulates dopamine, serotonin, and norepinephrine—neurotransmitters that govern focus and emotional balance.
Animal lab experiments demonstrated that regular caffeine intake reduces the production of proinflammatory cytokines IL-1β, TNF-α, and IL-6, preventing microglial cell damage during chronic stress.
Additionally, caffeine maintains levels of the BDNF protein (brain-derived neurotrophic factor) in the hippocampus, which is responsible for neuroplasticity and memory. Decaffeinated beverages failed to show a similar neuroprotective effect.
Human Study Data and Overdose Risks
According to a population study in Scientific Reports involving a sample of 1,994 people, drinking at least one cup of coffee daily was associated with a moderate reduction in the frequency of depressive states (odds ratio 0.70).
However, researchers stress that caffeine is not an antidepressant. Its impact on the body is strictly non-linear:
- Increased anxiety: high doses trigger panic attacks, particularly in people with variations of the ADORA2A gene;
- Disrupted sleep architecture: evening intake blocks melatonin production and deep sleep stages, which in itself worsens burnout;
- Tolerance: constant stimulation reduces receptor sensitivity, while sudden withdrawal causes withdrawal syndrome with headaches and irritability.
Experts advise viewing caffeine as a functional modulator of psychomotor responses and alertness that requires individual moderation.
